BODIPY 480/508-Cholesterol（簡稱：Bdp-Chol），CAS NO：878557-19-8，是一種具生物活性和細胞膜滲透性的膽固醇類似物，在C24位偶聯(lián)了一個BODIPY熒光基團。HeLa細胞中，Bdp-Chol與膽固醇標(biāo)記物-脫氫麥角甾醇（Dehydroergosterol, DHE）共定位；BHK細胞中，Bdp-Chol 從質(zhì)膜運輸進入內(nèi)吞再循環(huán)小泡（ERC）。Bdp-Chol最大激發(fā)和發(fā)射波長分別是480nm和508nm，用于監(jiān)測細胞內(nèi)的固醇攝取和細胞器間固醇通量。

產(chǎn)品使用：

以下是針對細胞樣本的染色方法，僅作參考。用戶需根據(jù)實際染色樣本和染色條件來優(yōu)化。

1、儲存液的制備

將低溫保存的固體平衡至室溫至少20min，低速離心后，于無菌工作臺內(nèi)加入適量有機溶劑（比如：無水高質(zhì)量DMSO）配置成1mM儲存液，根據(jù)單次用量分裝，充入惰性氣體后，≤-20℃避光凍存。

2、工作液的制備

于正式實驗前，用PBS、HBSS、無血清培養(yǎng)基等稀釋儲存液，到所需的工作濃度，比如：0.1-1μM。最佳的工作濃度請參閱文獻或自行設(shè)置梯度濃度進行摸索。工作液必須現(xiàn)配現(xiàn)用。

應(yīng)用示例（來自文獻，僅做參考）

用BODIPY-Cholesterol進行染色，方法如下：BODIPY-Cholesterol溶于DMSO配置1mM儲存液，分裝到玻璃瓶內(nèi)，充氮氣，置于-80℃凍存。精子懸液用BODIPY-Cholesterol（0.4 μM）標(biāo)記，混勻后，37℃孵育10min。通過二步法不連續(xù)梯度等密度Percoll去除多余的染料。精子調(diào)整密度到1×10⁶sperm/mL，孵育2h后進行流式評估。
